The son of John and Sarah (St. John) Gregory, he married Mary (maiden name unknown) in 1669. In 1666 he was granted a four acre homelot on the east side of Town Street. He had an estate of pounds in 1671 with two children and in 1687 it was valued at 100 pounds. He and his brother, Judah were listed as freemen of Norwalk in 1669. He served as fence viewer in 1674/5, overseer of pounds in 1679, and deputy at the General Court in 1695. He inherited land from his father in 1589. In 1697, he was part of a group of inhabitants who petitioned the court to purchase land north of Norwalk to create a plantation (north Redding). The inventory of his estate was taken in 1697/8 by his brother, John Gregory and brother-in-law John Benedict.

His widow, Mary and sons, Samuel and Thomas were appointed administrators of his estate which amounted to 415 pounds, 2 shilllings, 10 pence. The sons of Jachin Gregory, deceased, (Thomas, Matthew and Jachin, Jr.) and son-in-law, John Seymour, signed an agreement concerning his estate. Their brother, Samuel was deceased.
